    Oxidative stress and fetal growth restriction set up earlier in undernourished sheep twin pregnancies prevention with antioxidant and nutritional supplementation2022-06-28

    Hypoxemia and oxidative stress, resulting in intrauterine growth restriction (IUGR) in undernourished twin sheep pregnancies, has been described in near-term studies. Our aim was to evaluate if the counteractive effects of maternal nutritional or antioxidant supplementation on the fetal redox status were evident before the accelerated fetal growth phase. Forty twin-bearing ewes grazing on natural Patagonian prairie were randomly assigned to four groups (n = 10 each; P: control ewes consuming mainly natural pasture; P+A: pasture plus antioxidants; P+C: pasture plus concentrate; P+A+C: pasture plus antioxidants and concentrate). Daily herbal antioxidants were supplemented in a feedstuff concentrate as a premix from day 35 until day 100 of gestation, when fetal venous cord blood samples and biometric measurements were obtained via cesarean section. The fetuses from group P were clearly hypoxemic. An analysis of variance showed that maternal antioxidant supplementation showed a trend of increased PO2, SatHb, and Ht, effects not observed in P+C fetuses. Antioxidants decreased the fetal MDA concentration (p < 0.05). Fetal TAC was increased by the antioxidants and concentrate (p < 0.05). Antioxidant supplementation showed a trend to increase fetal body weight but not biometry. The results suggest that negative effects of oxidative stress occur earlier than the overt growth arrest, and the maternal administration of antioxidants may constitute a good nutritional strategy for the early prevention of IUGR.

    Antioxidant capacity of 5-Fluorouracile and new fluorinated uracil derivates2018-03-01

    Oxidative stress is associated with multiple pathologies such as cancer and can exacerbate the development of them. In this work, we have studied the antioxidant capacity of 5-Fluorouracile (5-FU) which is an antineoplastic drug that is used in the treatment of colorectal cancer. 5-FU is a compound that has a chemical structure similar to uracil and is also fluorinated. New fluorinated derivates previously obtained in our laboratory were tested to study its antioxidant activity. All the compounds analyzed were able to inhibit lipid peroxidation when used in concentrations of 10 μM.

    Effect of copaiba essential oil (Copaifera officinalis L.) as a natural preservative on the oxidation and shelf life of sheep burgers2021-03-18

    We evaluated the effects of the inclusion of copaı´ba (Copaifera officinalis L.) essential oil at 0.05 and 0.1% as a possible replacement of synthetic additive butylated hydroxytoluene (BHT) in sheep burgers during 14 days of storage in a refrigerated display case (4˚C). During the shelf life days, analyzes of antioxidant activity, lipid oxidation, pH, color, cooking loss, texture and consumer acceptability were carried out on refrigerated burgers. The addition of copaı´ba essential oil showed an antioxidant effect in sheep burgers (P > 0.05), reducing lipid oxidation. Copaiba essential oil added at 0.05% showed the highest antioxidant activity, decreased cooking losses and delaying discoloration (loss of redness) during storage; it further improved, the tenderness of sheep burgers (P < 0.05). The treatments had no effect (P > 0.05) on consumer acceptability. Copaiba essential oil is a promising natural antioxidant to increase the shelf life of meat products, as well as being a viable solution to replace synthetic antioxidant BHT, thereby promoting the fresh-like quality appeal of sheep burgers.

    Maternal supplementation with polyphenols and omega-3 fatty acids during pregnancy : prenatal effects on growth and metabolism2021-06-07

    Maternal supplementation with antioxidants and n-3 PUFAs may be a promising strategy to reduce the risk of intrauterine growth restriction and preterm delivery, which may diminish the appearance of low-birth-neonates. A previous studies showed beneficial outcomes of the combination of hydroxytyrosol and linoleic acid, but there is no data of its prenatal effects. The present study aimed to determine the possible prenatal implications of such maternal supplementation at prenatal stages in swine, a model of IUGR pregnancies. Results showed effects on litter size, with treated sows having larger litters and, therefore, smaller fetuses. However, the brain/head weight ratio showed a positive effect of the treatment in development, as well as in some other major organs like lungs, spleen, or kidneys. On the other hand, treated piglets showed better glycemic and lipidemic profiles, which could explain postnatal effects. However, further research on the implications of the treatment on litter size and prenatal and postnatal development must be done before practical recommendation can be given.

    Supplementation with proline improves haemato-biochemical and reproductive indicators in male rabbits affected by environmental heat-stress2021-02-02

    Twenty-four adult rabbit bucks (n = 6 per treatment) were fed a basal diet supplemented with 0 (control), 50, 100, and 150 mg proline/kg dry matter (DM) diet for 12 weeks to determine possible usefulness for alleviating the negative impact of environmental heat stress on redox status, haemato-biochaemical attributes and semen quality. There were significant dose–response effects, with increments in levels of dietary proline (LDP) quadratically improving red blood cell counts (p = 0.017), rectal temperature (p = 0.009), and respiratory rate (p < 0.001). Increasing LDP cubically affected superoxide dismutase activity in blood plasma (p = 0.012) and total antioxidant capacity in both blood and seminal plasma (p < 0.001 and p = 0.006, respectively). The optimal response was observed at 30 and 80 mg proline/kg DM for blood and seminal plasma, respectively. With regards to homeostasis indexes, increments in LDP cubically modified blood plasma concentrations of total protein (p = 0.002) and albumin (p < 0.001), with an optimal response found at 70 mg proline/kg DM. A linear relationship (p = 0.005) was also observed between LDP and blood plasma glucose concentrations, with the optimal response being found at 100 mg proline/kg DM. Increasing LDP also showed positive effects on reproductive traits, with quadratic increases in blood plasma testosterone and cortisol concentrations (p < 0.001; optimal responses at 50 and 60 mg proline/kg DM, respectively), a positive linear relationship with in libido, ejaculate volume, sperm concentration and total sperm count (p < 0.001 for all; optimal responses observed at 100 mg proline/kg DM) and a quadratic increase in total functional sperm fraction (p < 0.001; optimal response at 70 mg proline mg/kg DM). Hence, the optimal positive effects of dietary proline supplementation on redox status, blood metabolites, and reproductive traits of rabbit bucks may be achieved at 50–100 mg/kg DM.

    Organic and conventional tiger nut (Cyperus esculentus) : differences in nutritional, antioxidant and microbiological properties2020-10-01

    Tiger nut (Cyperus esculentus L.), also known as chufa (European sedge), is a member of the Cyperaceae family, which is used in organic and conventional agriculture for its small edible tubers and grown in temperate and tropical zones of the world being consumed raw, roasted or pressed for its juice as beverage. The aim of this study is analyzing the proximate composition (AOAC methods), total phenolic content (Folin-Ciocalteu method), total antioxidant capacity (Trolox equivalent antioxidant capacity, TEAC) and microbiological profile (International Standard Organization norms, ISO) of samples of Spanish organic and conventional, Nigerian conventional and unknown origin tiger nuts obtained from supermarkets and local markets in Spain. No significant differences in proximate composition and antioxidant properties were found between Spanish organic and conventional tiger-nuts, except in total phenolic level (p<0.005). No significant differences were found in all samples about ash, lipid, total sugar and carbohydrate values, but significant differences (p<0.005) were found for moisture, protein, fiber and antioxidant capacity between Spanish samples and Nigerian or unknown origin samples. No coagulase-positive staphylococci, nor Salmonella spp. were detected in any of the studied samples. Significant differences (p<0.005) were found for moulds and yeasts between analysed samples, but no significant differences were detected in other microorganisms.

    Polyphenols in farm animals : source of reproductive gain or waste?2020-10-21

    Reproduction is a complex process that is substantially a ected by environmental cues, specifically feed/diet and its components. Farm animals as herbivorous animals are exposed to a large amount of polyphenols present in their natural feeding system, in alternative feed resources (shrubs, trees, and agro-industrial byproducts), and in polyphenol-enriched additives. Such exposure has increased because of the well-known antioxidant properties of polyphenols. However, to date, the argumentation around the impacts of polyphenols on reproductive events is debatable. Accordingly, the intensive inclusion of polyphenols in the diets of breeding animals and in media for assisted reproductive techniques needs further investigation, avoiding any source of reproductive waste and achieving maximum benefits. This review illustrates recent findings connecting dietary polyphenols consumption from di erent sources (conventional and unconventional feeds) with the reproductive performance of farm animals, underpinned by the findings of in vitro studies in this field. This update will help in formulating proper diets, optimizing the introduction of new plant species, and feed additives for improving reproductive function, avoiding possible reproductive wastes and maximizing possible benefits.

    Oxygenic metabolism in nutritional obesity induced by olive oil : the influence of vitamin C2019-06-03

    Obesity is a medical and sociological problem of great importance due to the high percentage of people affected and the important health consequences that it involves. Most cases of obesity are related to an inadequate diet, rich in fats, which could lead to changes in the patient’s oxygenic metabolism. That is why this study has been proposed to evaluate how some aspects of oxygenic metabolism are affected in a nutritional experimental model, with a controlled hyperlipidic liquid diet based on olive oil, and the effect of the antioxidant vitamin C on these conditions. Wistar rats were divided into four groups which received a control and hyperlipidic liquid diet for 30 days, with or without a vitamin C supplement (CO, COC, HO and HOC). First of all the body and fat tissue development was measured in the four groups. Our results showed that the excessive intake of nutritional and healthy fat such as olive oil did not prevent the appearance of obesity and the supplementation with vitamin C did not have a protective effect on body and fat development. The study of the antioxidant enzymes superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) in total liver, liver cytosol, abdominal white fat, brown fat and blood cells showed that vitamin C could have different selectivities and affinities for different enzymes and compartments/tissues of the body. Finally, the effect of vitamin C on various metabolic parameters (glucose, pyruvate, lactate, LDH, ATP, acetoacetate and beta-hydroxybutyrate) provided positive protection against oxidative stress especially under hyperlipidic conditions. All things considered, the present study concludes that vitamin C treatment could protect Wistar rats from the oxidative stress impairment induced by obesity generated by an excessive intake of fats.

    Nitrosative stress in retinal pathologies : review2019-11-11

    Nitric oxide (NO) is a gas molecule with diverse physiological and cellular functions. In the eye, NO is used to maintain normal visual function as it is involved in photoreceptor light transduction. In addition, NO acts as a rapid vascular endothelial relaxant, is involved in the control of retinal blood flow under basal conditions and mediates the vasodilator responses of different substances such as acetylcholine, bradykinin, histamine, substance P or insulin. However, the retina is rich in polyunsaturated lipid membranes and is sensitive to the action of reactive oxygen and nitrogen species. Products generated from NO (i.e., dinitrogen trioxide (N2O3) and peroxynitrite) have great oxidative damaging ffects. Oxygen and nitrogen species can react with biomolecules (lipids, proteins and DNA), potentially leading to cell death, and this is particularly important in the retina. This review focuses on the role of NO in several ocular diseases, including diabetic retinopathy, retinitis pigmentosa, glaucoma or age-related macular degeneration (AMD).
